Description:
KLK11 Human Recombinant produced in HEK-293 cells is a single, glycosylated, polypeptide chain having a molecular weight of 25.6kDa though on SDS-PAGE migrates at about 41kDa due to the glycosilation.
The KLK11 is purified by proprietary chromatographic techniques.

Purity:
Greater than 95.0% as determined by SDS-PAGE.
Solubility:
It is recommended to reconstitute the lyophilized KLK11 in sterile 18MΩ–cm H2O not less than 100 µg/ml, which can then be further diluted to other aqueous solutions.
Formulation:
The KLK11 protein was lyophilized from a 0.2 µm filtered solution containing no additives or preservatives.
Stability:
Lyophilized KLK11 although stable at room temperature for 3 weeks, should be stored desiccated below -18 °C. Upon reconstitution KLK11 should be stored at 4 °C between 2-7 days and for future use below -18 °C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).

Activity:
KLK11 specific activity is greater than 2200 pmoles/min/µg when measure with 100uM colormetric peptide substrate (D-Val-Leu-Lys-ThioBenzyl ester) and 1 µg of activated enzyme. The reaction is carried out in a volume of 100µL containing 50mM Tris, 1M NaCl, 10mM EDTA, 0.1mM DTNB (5,5'Dithio-bis(2-nitrobenzoic acid)), pH8.5 at 37 °C. Cleavage of the peptide substrate can be measured at a wavelength of 405nm and quantified using the extinction coeffiecient 13,260M-1cm-1.

Sterile Filtered White lyophilized (freeze-dried) powder.
Activation:
KLK11 is activated in a reaction containing 100µg/ml Kallikrein 11, 10 µg/ml thermolysin, 50mM Tris, 10mM CaCl2, 150mM NaCl, 0.05% Brij 35, pH 7.5 for 15 minutes at 37 °C. The thermolysin reaction is stopped by adding EDTA to a final concentration of 50mM.
